    Vzorce a procesy prostorové distribuce rostlinných druhů napříč škálami

    In this thesis, I aimed to identify factors shaping plant distribution at different spatial scales, correlate them with environmental heterogeneity, identify causal processes and test general hypothesis on the nature of response curve shapes and species richness patterns. General review of the topic is introduced in the first chapter, followed by five chapters presenting three already published studies and two manuscripts. The first study deals with processes responsible for creation of fine-scaled spatial pattern of spruce seedlings and saplings, emerging after bark-beetle disturbance in mountain spruce forest. Aggregated pattern, replicating previous generation of spruce trees, emerges in consequence to microsite-dependent mortality, as was surveyed through repeated monitoring of the fate of individual seedlings. The second study explores spatial variability in forest understory temperatures at the landscape scale and its relevance for understory plant distribution. As the main source of variability in understory communities we identified seasonal maximum temperatures. Using GIS modelling approach, we created spatially continuous prediction, which outperformed state-of-art climatic grids used currently by ecologists. The third study on the shape of species responses along elevational gradients...SHRNUTÍ V této práci se zabývám procesy a faktory určujícími rozšíření rostlinných druhů na různých prostorových škálách. Studium prostorové distribuce má tři základní kroky: popis pozorovaného vzorce, nalezení prostorových vztahů s dalšími prostorovými jevy a nakonec určení kauzálních vztahů. Tato práce přináší pět případových studií, operujících na různých prostorových škálách, které přináší různou úroveň poznání prostorových vztahů a jejich příčin: V první studii se zabýváme na jemné škále identifikací procesů, které vedou ke vzniku shlukovitého rozmístění semenáčků smrku v porostech horských smrčin zasažených gradací kůrovce, které je navíc konzervativní v čase - kopíruje totiž předchozí generaci lesa. S pomocí dlouhodobého sledování jednotlivých semenáčků jsme identifikovali rozdíly v mortalitě na mikrostanovištích zodpovědné za prostorové shlukování semenáčků. Druhá studie zkoumá prostorovou variabilitu teploty v lesních porostech na krajinném měřítku a její vztah k rozšíření druhů bylinného patra. Jako hlavní složku teplotní variability zodpovědnou za rozšíření rostlin jsme identifikovali maximální teploty ve vegetační sezóně.     OPTIMIZATION OF 2‐STAGE TURBOCHARGED GAS SI ENGINE UNDER STEADY STATE OPERATION

    The proposed paper deals with an optimization of a highly‐turbocharged large‐bore gas SI engine. Only steady state operation (constant engine speed and load) is considered. The paper is mainly focused on theoretical potential of 2‐stage turbocharging concept in terms of performance and limitation. The results are obtained by means of simulation using complex 0‐D/1‐D engine model including the control algorithm. Different mixture composition concepts are considered to satisfy different levels of NOx limit – fresh air mixed with external cooled EGR is supposed to be the right approach while optimal EGR level is to be found. Considering EGR circuit, 5 different layouts are tested to select the best design. As the engine control is relatively complex (2‐sage turbocharger group, external EGR, compressor blow‐by, controlled air excess), 5 different control means of boost pressure were considered. Each variant based on above mentioned options is optimized in terms of compressor/turbine size (swallowing capacity) to obtain the best possible BSFC. The optimal variants are compared and general conclusions are drawn. Práce se zabývá optimalizací vysoce přeplňovaného velkého plynového zážehového motoru. Pouze ustálené režimy (konstantní otáčky a zátěž) jsou uvažovány. Článek je primárně zaměřen na teoretický potenciál dvoustupňového přeplňování z hlediska dosažených parametrů a omezení. Výsledky byly získány pomocí komplexního 0‐D/1‐D modelu motoru včetně řídícího algoritmu. Různé koncepce složení směsi jsou uvažovány pro dosažení různých limitů pro NOX – předpokládá se, že správný přístup zahrnuje mísení čerstvé směsi s EGR a hledá se optimální úroveň EGR. Co se týká EGR, tak je testováno celkem 5 variant okruhů EGR, ze kterých se vybírá nejlepší varianta. Protože řízení motoru je relativně komplexní (dvoustupňová plnicí skupina, vnější EGR, přepouštění na straně kompresoru, přebytek vzduchu), je testováno 5 různých způsobů řízení plnicího tlaku. Každá varianta založena na výše uvedených konfiguracích je optimalizována z hlediska velikosti kompresoru/turbiny (hltnost) pro nejlepší měrnou spotřebu.     Habitat preferences and functional traits drive longevity in Himalayan high-mountain plants

    Plant lifespan has important evolutionary, physiological, and ecological implications related to population persistence, community stability, and resilience to ongoing environmental change impacts. Although biologists have long been puzzled over the extraordinary variation in plant lifespan and its causes, our understanding of interspecific variability in plant lifespan and the key internal and external factors influencing longevity remains limited. Here, we demonstrate the concurrent impacts of environmental, morphological, physiological, and anatomical constraints on interspecific variation in longevity among > 300 vascular dicot plant species naturally occurring at an elevation gradient (2800–6150 m) in the western Himalayas. First, we show that plant longevity (ranging from 1 to 100 years) is largely related to species' habitat preferences. Ecologically stressful habitats such as alpine and subnival host long-lived species, while productive ruderal and wetland habitats contain a higher proportion of shorter-lived species. Second, longevity is influenced by growth form with monocarpic forbs having the shortest lifespan and woody shrubs having the highest. Small-statured cushion plants with compact canopies and deep roots, most found on cold and infertile alpine and subnival soils, had a higher chance of achieving longevity. Third, plant traits reflecting plant adaptations to stress and disturbance affect interspecific differences in plant longevity. We show that longevity and growth are negatively correlated. Slow-growing species are those that have a higher chance of reaching a high age. Finally, higher longevity was associated with high leaf carbon and phosphorus, low root phosphorus and nitrogen, and with large bark-xylem ratio. Our findings suggest that plant longevity in high elevation is intricately determined by a combination of habitat preferences and growth form, as well as the plant growth rate and physiological processes.publishedVersio

    Antibody expressing pea seeds as fodder for prevention of gastrointestinal parasitic infections in chickens

    <p>Abstract</p> <p>Background</p> <p>Coccidiosis caused by protozoans of genus <it>Eimeria </it>is a chicken parasitic disease of great economical importance. Conventional disease control strategies depend on vaccination and prophylactic use of anticoccidial drugs. Alternative solution to prevent and treat coccidiosis could be provided by passive immunization using orally delivered neutralizing antibodies. We investigated the possibility to mitigate the parasitic infection by feeding poultry with antibody expressing transgenic crop seeds.</p> <p>Results</p> <p>Using the phage display antibody library, we generated a panel of anti-<it>Eimeria </it>scFv antibody fragments with high sporozoite-neutralizing activity. These antibodies were expressed either transiently in agrobacteria-infiltrated tobacco leaves or stably in seeds of transgenic pea plants. Comparison of the scFv antibodies purified either from tobacco leaves or from the pea seeds demonstrated no difference in their antigen-binding activity and molecular form compositions. Force-feeding experiments demonstrated that oral delivery of flour prepared from the transgenic pea seeds had higher parasite neutralizing activity <it>in vivo </it>than the purified antibody fragments isolated from tobacco. The pea seed content was found to protect antibodies against degradation by gastrointestinal proteases (>100-fold gain in stability). <it>Ad libitum </it>feeding of chickens demonstrated that the transgenic seeds were well consumed and not shunned. Furthermore, feeding poultry with shred prepared from the antibody expressing pea seeds led to significant mitigation of infection caused both by high and low challenge doses of <it>Eimeria </it>oocysts.</p> <p>Conclusion</p> <p>The results suggest that our strategy offers a general approach to control parasitic infections in production animals using cost-effective antibody expression in crop seeds affordable for the animal health market.</p

    Fluvastatin in the therapy of acute coronary syndrome: Rationale and design of a multicenter, randomized, double-blind, placebo-controlled trial (The FACS Trial)[ISRCTN81331696]

    BACKGROUND: Activation of inflammatory pathways plays an important contributory role in coronary plaque instability and subsequent rupture, which can lead to the development of acute coronary syndrome (ACS). Elevated levels of serum inflammatory markers such as C-reactive protein (CRP) represent independent risk factors for further cardiovascular events. Recent evidence indicates that in addition to lowering cholesterol levels, statins also decrease levels of inflammatory markers. Previous controlled clinical trials reporting the positive effects of statins in participants with ACS were designed for very early secondary prevention. To our knowledge, no controlled trials have evaluated the potential benefits of statin therapy, beginning immediately at the time of hospital admission. A previous pilot study performed by our group focused on early initiation of cerivastatin therapy. We demonstrated a highly significant reduction in levels of inflammatory markers (CRP and interleukin-6). Based on these preliminary findings, we are conducting a clinical trial to evaluate the efficacy of another statin, fluvastatin, as an early intervention in patients with ACS. METHODS: The FACS-trial (Fluvastatin in the therapy of Acute Coronary Syndrome) is a multicenter, randomized, double-blind, placebo-controlled study evaluating the effects of fluvastatin therapy initiated at the time of hospital admission. The study will enroll 1,000 participants admitted to hospital for ACS (both with and without ST elevation). The primary endpoint for the study is the influence of fluvastatin therapy on levels of inflammatory markers (CRP and interleukin-6) and on pregnancy associated plasma protein A (PAPP-A). A combined secondary endpoint is 30-day and one-year occurrence of death, nonfatal myocardial infarction, recurrent symptomatic ischemia, urgent revascularization, and cardiac arrest. CONCLUSION: The primary objective of the FACS trial is to demonstrate that statin therapy, when started immediately after hospital admission for ACS, results in reduction of inflammation and improvement of prognosis. This study may contribute to new knowledge regarding therapeutic strategies for patients suffering from ACS and may offer additional clinical indications for the use of statins

    Unmasking Differential Effects of Rosiglitazone and Pioglitazone in the Combination Treatment with n-3 Fatty Acids in Mice Fed a High-Fat Diet

    Combining pharmacological treatments and life style interventions is necessary for effective therapy of major diseases associated with obesity, which are clustered in the metabolic syndrome. Acting via multiple mechanisms, combination treatments may reduce dose requirements and, therefore, lower the risk of adverse side effects, which are usually associated with long-term pharmacological interventions. Our previous study in mice fed high-fat diet indicated additivity in preservation of insulin sensitivity and in amelioration of major metabolic syndrome phenotypes by the combination treatment using n-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFA) and rosiglitazone, i.e. an anti-diabetic drug of the thiazolidinedione (TZD) family. We investigated here whether pioglitazone, a TZD-drug in clinical use, could elicit the additive beneficial effects when combined with n-3 LC-PUFA. Adult male mice (C57BL/6N) were fed an obesogenic corn oil-based high-fat diet (cHF) for 8 weeks, or randomly assigned to various dietary treatments (i) cHF+F, cHF with n-3 LC-PUFA concentrate replacing 15% of dietary lipids; (ii) cHF+ROSI, cHF with 10 mg rosiglitazone/kg diet; (iii) cHF+F+ROSI; (iv) cHF+PIO, cHF with 50 mg pioglitazone/kg diet; and (v) cHF+F+PIO, or chow-fed. Plasma concentrations of 163 metabolites were evaluated using a targeted metabolomics approach. Both TZDs preserved glucose homeostasis and normal plasma lipid levels while inducing adiponectin, with pioglitazone showing better effectiveness. The beneficial effects of TZDs were further augmented by the combination treatments. cHF+F+ROSI but not cHF+F+PIO counteracted development of obesity, in correlation with inducibility of fatty acid β-oxidation, as revealed by the metabolomic analysis. By contrast, only cHF+F+PIO eliminated hepatic steatosis and this treatment also reversed insulin resistance in dietary obese mice. Our results reveal differential effects of rosiglitazone and pioglitazone, unmasked in the combination treatment with n-3 LC-PUFA, and support the notion that n-3 LC-PUFA could be used as add-on treatment to TZDs in order to improve diabetic patient's therapy

    Fluvastatin in the first-line therapy of acute coronary syndrome: results of the multicenter, randomized, double-blind, placebo-controlled trial (the FACS-trial)

    <p>Abstract</p> <p>Background</p> <p>Statins have been proved to be effective in reduction of mortality and morbidity when started in the early secondary prevention in stabilized patients after acute coronary syndrome (ACS). The safety and efficacy of statin administration directly in the first-line therapy in unstable ACS patients is not clear. The aim of our study was, therefore, to assess the effect of statin treatment initiated immediately at hospital admission of patients with ACS.</p> <p>Methods</p> <p>The trial was stopped prematurely after enrollment of one hundred and fifty-six patients with ACS that were randomized at admission to fluvastatin 80 mg (N = 78) or placebo (N = 78). Study medication was administered immediately after randomization and then once daily for 30 days; all patients were then encouraged to continue in open-label statin therapy and at the end of one-year follow-up 75% in the fluvastatin group and 78% in the placebo group were on statin therapy.</p> <p>Results</p> <p>We did not demonstrate any difference between groups in the level of C-reactive protein, interleukin 6, and pregnancy-associated plasma protein A on Day 2 and Day 30 (primary endpoint). Fluvastatin-therapy, however, significantly reduced one-year occurrence of major adverse cardiovascular events (11.5% vs. 24.4%, odds ratio (OR) 0.40, 95% CI 0.17-0.95, P = 0.038). This difference was caused mainly by reduction of recurrent symptomatic ischemia (7.7% vs. 20.5%, OR 0.32, 95% CI 0.12-0.88, P = 0.037).</p> <p>Conclusions</p> <p>This study failed to prove the effect of fluvastatin given as first-line therapy of ACS on serum markers of inflammation and plaque instability. Fluvastatin therapy was, however, safe and it may reduce cardiovascular event rate that supports immediate use of a statin in patients admitted for ACS.</p> <p>Trial registration</p> <p>NCT00171275</p

    Responses of competitive understorey species to spatial environmental gradients inaccurately explain temporal changes

    Understorey plant communities play a key role in the functioning of forest ecosystems. Under favourable environmental conditions, competitive understorey species may develop high abundances and influence important ecosystem processes such as tree regeneration. Thus, understanding and predicting the response of competitive understorey species as a function of changing environmental conditions is important for forest managers. In the absence of sufficient temporal data to quantify actual vegetation changes, space-for-time (SFT) substitution is often used, i.e. studies that use environmental gradients across space to infer vegetation responses to environmental change over time. Here we assess the validity of such SFT approaches and analysed 36 resurvey studies from ancient forests with low levels of recent disturbances across temperate Europe to assess how six competitive understorey plant species respond to gradients of overstorey cover, soil conditions, atmospheric N deposition and climatic conditions over space and time. The combination of historical and contemporary surveys allows (i) to test if observed contemporary patterns across space are consistent at the time of the historical survey, and, crucially, (ii) to assess whether changes in abundance over time given recorded environmental change match expectations from patterns recorded along environmental gradients in space. We found consistent spatial relationships at the two periods: local variation in soil variables and overstorey cover were the best predictors of individual species’ cover while interregional variation in coarse-scale variables, i.e. N deposition and climate, was less important. However, we found that our SFT approach could not accurately explain the large variation in abundance changes over time. We thus recommend to be cautious when using SFT substitution to infer species responses to temporal changes.</p
